Kidney regeneration in fish.

چکیده

BACKGROUND Chronic and acute kidney injury damages nephrons, the blood filtering tubules in the kidney. Although mammalian kidneys can regenerate the tubular epithelium of the nephron, no new nephrons are made during adulthood. In contrast, fish are capable of growing nephrons de novo throughout their life. A better understanding of this 'neo-nephrogenic' response in fish may lead to the development of novel regenerative therapies to treat kidney disease in humans. SUMMARY In this review, nephron formation in the fish mesonephric kidney during normal growth and in response to acute injury is examined at the morphological and molecular levels. Included is an overview of the recent discovery of migratory nephron progenitors that, following transplantation, can engraft host kidneys and give rise to functional nephrons. KEY MESSAGES Mesonephric nephron progenitors appear during the larval stage, migrate together to form clusters, activate the expression of conserved nephrogenic genes, and epithelialize into nascent nephrons in a process that resembles mammalian nephron formation. Nephron progenitors persist in the adult fish kidney and continue to add new nephrons at a basal rate as the fish grows in size. Following acute kidney injury, nephron formation is significantly increased, allowing the fish to rapidly regenerate lost nephrons. Transplantation of nephron progenitors into the kidney results in the formation of donor-derived nephrons in the recipient fish.

A fish model of renal regeneration and development.

The fish kidney provides a unique model for investigating renal injury, repair, and development. Like mammalian kidneys, fish kidneys have the remarkable ability to repair injured nephrons, designated renal regeneration. This response is marked by a recovery from acute renal failure by replacing the injured cells with new epithelial cells, restoring tubule integrity. In addition, fish have the ...

Gentamicin-induced nephrotoxicity and nephroneogenesis in Oreochromis nilotica, a tilapian fish

The renal response to 2 doses of gentamicin (low dose of 5 mg kg-' of body weight and high dose of 25 mg kg-' of body weight) was examined in the tilapian fish Oreochromis nilotica. Gentamicin exposure induced acute tubular necrosis that peaked in severity at 2 d following intraperitonea] injection of the high dose and at 4 to 7 d following injection of the low dose. Necrosis following high dos...

Identification of progenitors capable of kidney regeneration in zebrafish

Unlike adult mammals, which can repair but not regenerate new nephrons, fish generate nephrons de novo throughout life and after kidney injury. To explore mechanisms of kidney regeneration that could one day be applied to treat renal disease in humans, Diep et al. used the zebrafish as a model to identify and characterise progenitor cells responsible for nephron regeneration. Using transplantat...

Bioaccumulation and Depuration of Copper in the Kidney and Liver of a Freshwater Fish, Capoeta fusca

Background: This study aims to investigate the patterns of bioaccumulation and depuration of copper in the selected kidney and liver of Capoeta fusca. Methods: The fish were collected between September and November 2010 from a qanat in Birjand. They were exposed to two types treatments with copper (0.25 and 0.75 mg/L) for a period of 41 days. The fish under study were exposed to the above-ment...

Histological analysis of acute toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in zebrafish.

Previous studies have demonstrated that acute exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) by injection leads to inhibition of caudal fin regeneration in zebrafish. Since the TCDD exposure in these studies is systemic, it is possible that pathology in organs other than the fin could result in inhibition of fin regeneration. Therefore, histopathology of adult zebrafish (Danio rerio) or...
